CURRENT PROJECTS

2- Computational Study of Novel Dual-Targeting Probe for Prostate Cancer Molecular Imaging

Prostate specific membrane antigen (PSMA) is a transmembrane protein often overexpressed in prostate cancer (PC).   SPSMA cleaves vitamin B9 to stimulate PI3K-Akt signaling through metabotropic glutamate receptors (mGluR). We computational study to design and synthesize novel Cy3-congugated diagnostic glutamate-urea-based probes (GUL-Cy3) for detecting PC with the dual-targeting to both   PSMA and mGluRs.

4- Rational Design of Altered Peptide Ligands for Treatment of Autoimmune Disorders

Celiac disease (CD) is an MHC type II autoimmune disorder affecting approximately 1% of western populations and is caused by gluten ingestion which leads to intestinal damage.1 Currently there is no cure. We propose the targeting of human leukocyte antigen (HLA) DQ2 receptors through the use of altered peptide ligands (APLs), which have been rationally designed and are able to competitively inhibit gluten binding and presentation to T cells, especially when combined with other approaches such as cyclization.

3- Computationally analyzing the interactions between the minor cannabinoids and their biological targets

There are 117 minor and understudied cannabinoids may prove even more potent. we attempted to computation study and investigation of the interactions of major, minor and endo- cannabinoids with CB1 and CB2, Serotonin 5-HTIA, Capsaicin, PPAR-Y, GABA-A and Fatty-Acid Binding Proteins with an emphasis on the published crystal structures of the these receptors. In-silico screening will explores the analysis of cannabinoid interactions with mentioned receptors in order to highlight key interactions which may provide clues to better understand the pharmacological effects of cannabis constituents on these receptors.

5- Study of Pholymorphysm in Spin Cross Over (SCO) Compounds

spin-crossover (SCO) materials are particularly attractive for potential applications in molecular sensing, switching, data storage, display, and other electronic devices at nanometric scale. Polymorphism is widely encountered in the studies of crystallization, phase transition, materials synthesis, biomineralization, and in the manufacture of drugs. Studying polymorphism of SCO compounds is therefore important for better understanding the structural factors contributing to spin transition and the structure–function relationship.

1- Repurposing and computationally and structurally some candidate drugs including integrase inhibitors with potential anti-SARS-CoV-2 potential.

Cotinga has a number of HIV integrase inhibitors in preclinical development and we will be using these same techniques to screen them against these proteins. Very preliminary computational results from our lab are predicting that these are far more potent inhibitors of the main protease than the commercial integrase inhibitors (based on induced docking scores and MM-PBSA energy analyses of molecular dynamics simulations).
